The effect of ethanol extract from pomegranate (Punica granatum) seeds on growth inhibition and expression of inducible nitric oxide synthase (iNOS) on HCT116 colorectal cancer cell line

Abstract

Purpose: To investigate the cytotoxic activity of ethanol extract from pomegranate seeds on growth inhibition and iNOS expression on HCT116 colorectal cancer cell line Methodology: Ethanol extract from Punica granotum seeds was prepared for maceration in ethanol. The in vitro anticancer activity of ethanol extract from Punica granatum seeds was tested with MTT assay. Cisplatin was used as control. The log concentration of extract were used to calculate IC50. The effect of ethanol extract from Punico gronotum seeds on decreasing expression of iNOS also studied by calculating the H-score from immunocytochemistry. H-score is semiquantitative scoring system, ranged from 0 to 300. It used in this study to asses the expression of iNOS protein on HCT116 colorectal cancer cell line. If H-score is > 200, it indicates that iNOS protein was expressed in high amount. If H-score is <200, it indicates that iNOS protein was expressed in low amount. There were four group in immunocytochemistry; negative control (0 ppm), small dose (50 ppm), medium dose (100 ppm), and large dose (200 ppm). sults: Ethanol extract from Punica granatum seeds showed anticancer activity with IC50 of 54.2763 jUg/ml. Ekspression of iNOS protein was decreased with mean H-score of 158.48 in large dose group (200 ppm) after administration of ethanol extract from Punica granatum seeds. alue: This study showed that ethanol extract from Punica granatum seeds has potency to be developed as chemopreventive agents.
