Isolation and screening of extracellular enzymes producing bacteria from black soldier fly larvae

Abstract

Hermetia illucens L. is known as Black Soldier Fly (BSF), a voracious feeders during larval stages. Its role as bioconversion of organic waste is assumed correlate with the bacteria in BSF larvae. The bacteria are expected to have enzyme activity such as amylase, lipase, and protease. In the present study, the bacteria from BSF larvae were isolated using 6 different media. The isolated bacteria will be screened for their amylolytic, lipolytic, and proteolytic activities. Based on the formation of a clear zone and good growth appearance, total 22 representative bacterial isolates were obtained. Characterization of the isolates was consist of the morphological and biochemical tests. According to the result of the enzymatic test, five isolates have positive result on the amylolytic test, two isolates have a positive result on the lipolytic test, and eleven isolates have a positive result on the proteolytic test. Among those isolates, the B18 isolate has positive result on the three enzymatic tests. Molecular identification based on 16S rRNA showed that B18 was closely related to Bacillus paramycoides with 99.93% sequences similarity.
