title = "In-vitro formation of adduct 8-hydroxy-2′-deoxyguanosine DNA against 2′-deoxyguanosine with the influence of nickel (II), copper (I), and copper (II) metals",
abstract = "Determination of 8-hydroxy-2'-Deoxyguanosine in this research is a form of biomarker that can be used as an early detection of DNA damage due to long-term xenobiotic exposure can trigger cancer. In-vitro studies in this research by reacting one of the BASE DNA is 2'-deoxyguanosine which is presented with the metals Ni(II), Cu(I), and Cu(II) as xenobiotic compounds assisted by H2O2. The reaction is carried out at a temperature of 37°C for 24 hours in an incubator shaker. Variations in this study were at pH 7.4 and 8.4. The products of the reaction were analyzed using a reverse-phase HPLC instrument using phosphate buffer and methanol as motion phase with a ratio of 85:15. The formation of 8-OHdG from the interaction of 2'-deoxyguanosine with the three metals and the addition of H2O2 at pH 8.4, temperature 37°C for 24 hours is higher than the interaction at pH 7.4. In more alkaline pH causes hydrogen peroxide to decompose more thus increasing the formation of free radicals as the cause of the formation of 8-OHdG.",
